The ChemiScreen P2Y1 Purinergic Receptor Stable Cell Line is an engineered cell line for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This cell line enables assessment of ligand (e.g. ADP, ATP) based activation of P2RY1 GPCR activity via detection of increasing levels of intracellular calcium. The included cell line overexpresses P2RY1 alongside endogenous high levels of the promiscuous G protein, Gα15, coupling the receptor to the calcium signaling pathway. Upon activation of P2RY1, the resulting calcium activity can be fluorescently assayed. This stable modified cell line is provided as two vials of cryopreserved cells. The KILR SKBR3 cell pool is a stable, engineered target cell pool, which closely reflects the heterogeneous native protein expression of tumor environments, used to measure direct cell death mediated by immunooncology treatments (e.g. checkpoint modulators, bispecifics, CAR-T, etc.) via Antibody Dependent Cell-mediated Cytotoxicity (ADCC), Complement Dependent Cytotoxicity (CDC), and Antibody Dependent Cell Phagocytosis (ADCP). This product uses EFC technology. The cell pool is a heterogeneous population of SKBR3 cells that overexpress a ProLabel-tagged housekeeping gene. Upon effector cells-mediated cell lysis, ProLabel is released to the media, where EA in the detection kit produces an EFC signal proportional to the level of ProLabel released. 5 doses of 10 µg/kg total body weight of Neupogen (filgrastim) are administered on each of 5 consecutive days and two 240ug/kg total body weight doses of Mozobil(plerixafor) are also administered on the 4th and 5th day prior to collection. Leukapheresis is performed on the day of the 5th administration and the following day* using the Spectra Optia Apheresis System. Mononuclear cell-rich blood is continuously collected using ACD-A as an anticoagulant. Approximately 120mL of autologous plasma is combined with apheresed cells. The product is filled to deliver a nominal 290mL and is shipped in a 300mL transfer bag. Total nucleated cells (TNC) in our fresh Neupogen/Mozobil-mobilized leukapheresis products are composed of, on average, 1.3% CD34+ hematopoietic stem and progenitor cells (HSPCs). Recognizes a protein of 62 kDa, identified as PAX8. It is a member of the paired box (PAX) family of transcription factors. This nuclear protein is involved in thyroid follicular cell development and expression of thyroid-specific genes.
